Pelayaran pertama James Cook merupakan ekspedisi Tentera Laut Diraja British dan Persatuan Diraja gabungan ke selatan Lautan Pasifik di kapal HMS Endeavour, dari 1768 hingga 1771. Ia merupakan yang pertama daripada tiga pelayaran Pasifik yang James Cook menjadi komander. Tujuan ekspedisi pertama ini untuk memerhati transit Zuhrah 1769 merentasi Matahari (3–4 Jun tahun itu), dan mencari bukti Terra Australis Incognita yang diandaikan atau "tanah selatan yang belum ditemui".
